Reply from the Authors: The corrections provided by Waring et al. raise more questions than they resolve. Each of their reaction tubes, described in detail, contained 750 times 10 sup -12 mol 2-mercaptoethanol (2ME). Even if they had allowed the TMT reaction to go to completion, they would be measuring no more than 50 times 10 sup -12 mol methylmercaptoethanol (MME) by injecting 10 micro liter of 150 micro liter resuspended product into the HPLC. Furthermore, since the reaction was linear for twice the 30-minute incubation period over a five-fold range of enzyme activity, they had to detect one-tenth this amount of MME (5 times 10 sup -12 mol).
