方法:我们研究无关的18例六氯(7家族、零星11日)为阴性NKX2-1突变。六氯的诊断是基于儿童运动障碍的存在,主要表现为舞蹈病和其他主要神经功能。ADCY5分析是由whole-exome测序或Sanger测序。ADCY5和NKX2-1表达在成人大脑发育和人类大脑是死亡的脑组织的评估使用微阵列分析。
结果:c.1252C > T;p。R418W突变被发现2例(1家族、零星的)。的家庭情况下遗传变异影响的父亲,温和得多表示,可能由于低级体细胞镶嵌性。突变是新创的零星的情况。这些病例的临床表现以非突发性广义舞蹈病,以及肌张力障碍最严重影响,但没有面部肌纤维颤搐。我们观察到症状的重要进展ADCY5突变携带者,而六氯二次NKX2-1突变。临床课程的差异反映出大脑表达数据,显示增加ADCY5表达在纹状体在大脑发育,而NKX2-1显示了一个相反的趋势。
结论:我们的研究识别突变ADCY5,先前的基因与家族性运动障碍与面部肌纤维颤搐,作为家族和零星的六氯的原因。ADCY5遗传分析应该执行的情况下良性舞蹈病样的运动障碍患者即使没有面部肌纤维颤搐。
